The present invention is directed to membrane active poly (vinyl ester) polymers and compositions for targeted delivery of RNA interference (RNAi) polynucleotides to cells in vivo. An RNAi polynucleotide is attached to a poly (vinyl ester) polymer and the polymer is reversibly modified to allow targeted delivery in vivo. The membrane activity of poly (vinyl ester) provides for the transfer of RNAi polynucleotides from outside the cell to inside the cell.
