The present invention relates to a multilamellar vesicle preparation containing as component (A): a compound represented by formula (1) wherein R1 and R2 are an alkyl group having 5 - 21 carbon atoms or an alkenyl group having 5 - 21 carbon atoms carbon, R3 and R4 are a hydrogen atom, an alkyl group having 1 to 22 carbon atoms or an alkenyl group having 2 to 22 carbon atoms, z is an integer not less than 0, and x and y are an integer of 2 - 4； or a salt thereof, as component (B): a physiologically active substance, and as component (C): water. The multilamellar vesicle preparation according to the invention can be produced conveniently and is superior in stability.
