PROBLEM TO BE SOLVED: To improve a production efficiency of desired protein by examining treatment of root or leaf portions immersed into an infection liquid comprising Agrobacterium in a step of infecting with Agrobacterium a plant used in the production of the desired protein by a plant.SOLUTION: A production method of a protein by a plant comprises the steps of: cultivating a plant which may be infected with Agrobacterium by hydroponics method contacting the plant grown by the hydroponics method comprising roots and leaves with an infection liquid containing Agrobacterium having a polynucleotide encoding the desired protein to infect and cultivating the postinfectious plant to express the desired protein in the plant. The infection step is performed by regulating pressure in a state that at least one portion of the plant is immersed in the infection liquid comprising the Agrobacterium. The ratio of the root immersed in the infection liquid in the step is 50 mass% or less of the entire root.
