Disclosed herein are methods for producing induced T regulatory cells (iTregs), involving contacting naive CD4+CD25- T cells and/or naive CD8+CD25- T cells in vitro with (i) a polypeptide comprising an Ig-C domain from a B7-H4 protein (also known as VTCNl, B7x or B7S 1), wherein the polypeptide is not the full B7-H4 protein and does not include the B7-H4 protein IgV domain, (ii) anti-CD3 antibodies, (iii) anti-CD28 antibodies, and (iv) interleukin-2 (IL-2), wherein the contacting occurs for a time and under conditions sufficient to cause conversion of naive CD4+CD25- T cells and/or naive CD8+CD25- T cells into iTregs.La présente invention concerne des procédés de production de cellules T régulatrices induites (iTreg), comprenant la mise en contact de cellules T CD4+CD25- naïves et/ou de cellules T CD8+CD25- naïves in vitro avec (i) un polypeptide comprenant un domaine dIg-C dune protéine B7-H4 (également appelé VTCNl, B7x ou B7S 1), le polypeptide nétant pas la protéine B7-H4 complète et ne comprenant pas le domaine dIgV de la protéine B7-H4, (ii) des anticorps anti-CD3, (iii) des anticorps anti-CD28, et (iv) linterleukine-2 (IL-2), la mise en contact se produisant sur une durée et dans des conditions suffisantes pour provoquer la conversion des cellules T CD4+CD25- naïves et/ou des cellules T CD8+CD25- naïves en iTreg.
