A kind of culture medium and culture method for Antrodia cinnamomea culture is provided. Said culture medium contains cereal composition as a matrix, and nutrient solution containing carbon source, nitrogen source, and peptide and flavonoids as a physiological regulator. Said culture method is to graft Antrodia cinnamomea to the above-described sterilized culture medium and culture for a period of time at room temperature so that Antrodia cinnamomea will generate mycelium, fruiting body and fruiting body-like structure.
