PROBLEM TO BE SOLVED: To provide a method of replicating virus in a culture of sCLDK cells derived from Dog Kidney cells that are not adherence growth type but are suspension growth type cells, in a media free of animal-derived components.SOLUTION: The present invention relates to animal cells which can be infected by viruses and are adapted to growth in suspension in the medium free of animal-derived components, such as serum-free medium, and a method for the replication of viruses in cell culture using these cells. The present invention also relates to a method of reducing the amount of the serum substitutes in the medium to zero, after serially passaging and growing sCLDK cells in suspension in the media containing one or more serum substitutes, and a method of harvesting the virus from sCLDK cells which are suspension cultures by, thereafter, inoculating sCLDK cells with virus and infecting sCLDK cells with virus, and thereafter, growing sCLDK cells.【課題】動物由来成分を含まない培地で、付着増殖型でない浮遊増殖型細胞であるイヌ腎細胞由来のｓＣＬＤＫ細胞の培養物においてウイルスを複製する方法【解決手段】ウイルスが感染することができ、無血清培地等の動物由来成分を含まない培地において浮遊状態で増殖するように適合させた動物細胞でありこれらの細胞を用いた細胞培養物におけるウイルス複製の方法。ＣＬＤＫ細胞を１種又は複数の血清代替物を含む培地で浮遊状態で連続継代培養を行った後、前記培地中の前記血清代替物の量をゼロに迄減少させる方法、その後ウイルスを接種し、ｓＣＬＤＫ細胞に感染させ、その後増殖させて、浮遊培養物であるｓＣＬＤＫ細胞からウイルスを回収する方法。【選択図】なし
