The present invention relates to a polysaccharide fraction which is derived from ginseng and treated by enzymes to have reinforced immunity enhancement effects and a preparing method thereof. More specifically, the method includes the steps of: sequentially applying two or more enzymes including amylase and cellulase to ginseng in order to execute an enzyme treatment process and obtaining the polysaccharide fraction with the molecular weight of 10kDa or more from the hydrolyzate of the enzyme-treated ginseng. The present invention can prepare the polysaccharide fraction with excellent immune activity increasing effects in a cost-effective manner by through a composite enzyme treatment process including the amylase and cellulase.
