The instant disclosure provides a rubber tourniquet for increasing elongation, a rubber composition thereof and a manufacturing method thereof. The rubber composition includes 100 wt.% of isoprene rubber, 50 to 10 wt.% of inorganic filler, 2 to 5 wt.% of vulcanizing agent, 5 to 10 wt.% of organic filler, and 10 to 20 wt.% of plasticizer. The rubber composition does not include proteins. The rubber composition of the instant disclosure is used instead of natural latex for producing tourniquet, thereby avoiding the allergic reaction that may initiated by proteins in natural latex.
