The invention relates to a method and device for measuring fluorescence in a surface zone of a biological tissue, using a probe. A luminous mark that is detectable with a second sensor (11) is projected onto a zone of the surface of the biological tissue. The position of the image of this mark on the second sensor (11) is used to locate and determine the coordinates of the fluorescence detected with a first sensor (10), in an image of the zone of biological tissue produced in the visible spectrum. Thus, using the second sensor (11), the distance between the surface zone of the biological tissue and a fixed point in the coordinate system of the probe is determined. The distance thus determined is used to calculate the distance between the biological tissue and the fluorescence sensor (10), and to focus this fluorescence sensor (10) on the surface zone of the biological tissue.Linvention concerne un procédé et un dispositif de mesure de la fluorescence sur une zone en surface dun tissu biologique, à laide dune sonde. On projette, sur une zone de la surface du tissu biologique, une marque lumineuse détectable par un deuxième capteur (11). La position de limage de cette marque sur le deuxième capteur (11) est utilisée pour localiser et repérer la fluorescence détectée par un premier capteur (10), sur une image de la zone de tissu biologique réalisée dans le visible. On détermine aussi, à laide du deuxième capteur (11) la distance entre la zone en surface du tissu biologique et un point fixe dans le repère de la sonde. La distance ainsi déterminée est utilisée pour calculer la distance entre le tissu biologique et le capteur de fluorescence (10), et faire la mise au point de ce capteur de fluorescence (10) sur la zone en surface du tissu biologique.
