PROBLEM TO BE SOLVED: To provide a cultured periodontal ligament cell sheet of high engraftment properties which has satisfactory adhesiveness to a dental root surface.SOLUTION: A cultured periodontal ligament cell sheet is manufactured by culturing a cell under a particular condition on a cell culture support body in which a surface of a base material is coated by temperature responsive polymer whose upper limit or lower limit critical dissolution temperature in water is 0 to 80°C, (1) setting culture solution temperature to upper limit critical dissolution temperature or higher or lower limit critical dissolution temperature or lower, (2) bringing the cultured periodontal ligament cell sheet which was cultured into tight contact to a carrier, and (3) peeling the sheet directly together with the carrier. The cell sheet has a ligament-like microstructure, has extremely high engraftment on the dental root surface, a target cell can be transplanted at high density, and periodontal tissue can be positively reconstructed. Furthermore, cell sheets to be transplanted are laminated to allow the sheets to have three-dimensional polarity, by which an attachment apparatus can be reconstructed even more efficiently, and clinical application is strongly expected for intermediate periodontitis, serious periodontitis, gingival retraction or the like.COPYRIGHT: (C)2014,JPO&INPIT【課題】歯根表面への付着性が良好な高生着性培養歯根膜細胞シートを提供すること。【解決手段】水に対する上限もしくは下限臨界溶解温度が0~80℃である温度応答性ポリマーで基材表面を被覆した細胞培養支持体上で細胞を特定の条件下で培養し、(1)培養液温度を上限臨界溶解温度以上または下限臨界溶解温度以下とし、(2)培養した培養歯根膜細胞シートをキャリアに密着させ、(3)そのままキャリアと共に剥離することにより製造された培養歯根膜細胞シートは、靭帯様微小構造を有し、歯根表面への生着性が極めて高く、高密度に標的細胞を移植させられ、積極的な歯周組織の再建をはかれる。さらに、移植する細胞シートの積層化を行い3次元的な極性を持たせることにより、一層効率良く付着器官を再構築させることができ、中等度歯周炎、重度歯周炎、歯肉退縮等の臨床応用が強く期待される。【選択図】なし
