PROBLEM TO BE SOLVED: To construct liposome imitating asymmetry of a biological membrane, and to form a domain.SOLUTION: An oily solution phase 2 is disposed on an aqueous solution phase 1, a lipid monomolecular film 3 to be an outer layer lipid membrane is formed on an oil/water interface, and W/O drop 5 in which an inner layer lipid membrane 6 is formed is introduced into the oily liquid phase 2. By imparting a specific gravity difference between an aqueous solution configuring a drop 7 and the aqueous solution configuring the aqueous solution phase 1, the W/O drop 5 is shifted into the aqueous solution phase 1 by gravity, and the outer layer lipid membrane (lipid monomolecular film 3) is formed on the outer side of the inner layer lipid membrane 6. The liposome 9 to be produced is provided with an asymmetrical lipid 2-molecular film wherein the compositions of the inner layer lipid membrane 6 and the outer layer lipid membrane (lipid monomolecular film 3) are different and a micro domain structure wherein specified lipid is unevenly distributed inside the film surface of the asymmetrical lipid 2 molecular film is formed.COPYRIGHT: (C)2010,JPO&INPIT【課題】 生体膜の非対称性を模倣したリポソームを構築し、ドメイン形成を実現する。【解決手段】 水性溶液相1上に油性溶液相2を配するとともに、油水界面に外層脂質膜となる脂質単分子膜3を形成しておき、油性液体相2中に内層脂質膜6が形成されたW/O液滴5を導入する。液滴7を構成する水性溶液と水性溶液相1を構成する水性溶液に比重差を付与することで、重力によりW/O液滴5を水性溶液相1中に移行させ、内層脂質膜6の外側に外層脂質膜(脂質単分子膜3)を形成する。作製されるリポソーム9は、内層脂質膜6と外層脂質膜(脂質単分子膜3)の組成が異なる非対称性脂質2分子膜を備え、当該非対称性脂質2分子膜の膜面内に特定の脂質が偏在したミクロドメイン構造が形成されている。【選択図】 図1
