PROBLEM TO BE SOLVED: To provide a versatility method for selecting barley capable of selecting barley containing high in malt extract with high reliability.SOLUTION: The method for selecting barley containing high in malt extract includes: an amplification step in which an amplified DNA fragment is obtained by using an amplifying primer set with MWG655A sequence corresponding to a base sequence of a specific sequence and carrying out polymerase chain reaction using genomic DNA of barley to be tested as a template； and a digestion step in which digestion is processed the amplified DNA fragment by a restriction enzyme HinfI. The barley to be tested is selected as barley containing high in malt extract when the DNA fragment, which is cut by the restriction enzyme in a restriction enzyme treated object obtained by the digestion step, exists.【課題】麦芽エキスの高い大麦を信頼性よく選抜することが可能な汎用性ある大麦選抜方法を提供する。【解決手段】麦芽エキスの高い大麦を選抜するための方法であって、特定の配列の塩基配列に相当するＭＷＧ６５５Ａ配列を増幅可能なプライマーセットを用い、被検大麦のゲノムＤＮＡを鋳型としたポリメラーゼ連鎖反応を行って、増幅ＤＮＡ断片を得る増幅工程と、増幅ＤＮＡ断片を制限酵素ＨｉｎｆＩで消化処理する消化工程と、を含み、消化工程で得られた制限酵素処理物中に前記制限酵素により切断されたＤＮＡ断片が存在する場合に、該被検大麦を麦芽エキスの高い大麦として選抜する方法。【選択図】なし
