A method of suppressing an immune response is provided. This method involves administering to a subject in need of such treatment an isolated lymphoid tissue-derived inhibitory stromal cell (LSSC) in an amount effective to suppress the immune response in the subject. The present invention also relates to a method of isolating LSSCs comprising digesting lymph tissue fragments using a combination of enzyme mixture and agitation followed by collection of LSSCs.
