This application refers to microspheres comprising oligonucleotides for treatment of type 1 diabetes, said oligonucleotides comprise between about 30 weight percent and about 100 weight percent of the microspheres, based on the total weight of the microspheres, said microspheres having an average particle size of not greater than about 50 microns. The oligonucleotides are targeted to bind to primary transcripts selected from the group consisting of CD40, CD80 and CD86 primary transcripts, and combinations thereof.Selon cette invention, des oligonucléotides antisens sont administrés sous forme de microsphères afin dinduire la tolérance de cellules dendritiques, plus spécifiquement dans le modèle de souris non obèse diabétique (NOD).
