A method for characterizing the radiosensitivity of a cell sample of a patient to ionizing radiation, said cell sample having been obtained from cells taken from a patient, wherein the method comprises: (a) isolating target nucleated cells cell sample; (b) irradiating said cell sample before or after step (a) with an absorbed dose D of ionizing radiation; (c) recovering cytoplasmic and nuclear fractions from the cell sample; (d) determining, on the cytoplasmic and / or nuclear fractions of said cell sample, either: (d1) the amount of substrate phosphorylated by the activated ATM protein at at least one observation time t between t1 times 1, 2 and t3 , preferably at least one observation time t2 after irradiation with an absorbed dose D, (d2) the kinase activity by quantifying pATM phosphorylated at at least one observation time t selected from t = t1, t2; (e) determining at least one parameter selected from the group consisting of: the grade of severity of the post-radiotherapy tissue reaction of the patient according to the CTCAE classification, using at least the average number pATMmax nuc (t); radiosensitivity of the cell sample using at least the average number pATM maxnuc (t); and where t1 is a fixed value which represents the time at which the number of recognized double-strand breaks (DSBs) peaks in control cells from radiation-resistant patients; • t2 is a fixed value representing the time at which approximately 50% of CBDs are repaired in control fibroblasts from radiation-resistant patients and the time after which 30% of CBDs are repaired in control lymphocytes or lymphoblasts from radiation-resistant patients ; T3 is a fixed value which represents z times t2, where z is between 1.1 and 8, and preferably between 1, 2 and 6.
