Provided herein are methods and compositions for treating cancer by modulating the microbiome to enhance the effectiveness of immune checkpoint blockade. The microbiome can be modulated by administering butyric acid and / or butyric acid producing bacteria. Also provided herein is a method of determining a response to an immune checkpoint inhibitor by identifying whether a subject has a favorable microbial profile.
