PROBLEM TO BE SOLVED: To provide variant Fc polypeptides that bind to the neonatal Fc receptor with a high affinity/high binding activity, as well as methods of isolation, preparation and use thereof.SOLUTION: The invention provides a variant Fc polypeptide comprising a human IgG1, IgG2, IgG3 or IgG4 variant Fc fragment that contains an insertion of 10-20 amino acids within or adjacent to a specific sequence, where the insertion comprises at least one cysteine in the first four inserted amino acids and at least one cysteine in the last four inserted amino acids, where the variant Fc polypeptide binds to human neonatal Fc receptor (hFcRn) with a higher binding activity when compared to a control Fc polypeptide having the same amino acid sequence except for not containing the insertion, and the variant Fc polypeptide has little or no binding activity to hFcRn at a certain condition with a residual binding response detected higher than that of the control Fc polypeptide by no more than 0.1 nanometer.SELECTED DRAWING: Figure 1【課題】高い親和力・大きな結合活性で新生児Ｆｃ受容体に結合する変異型Ｆｃポリペプチドとその単離・作製・使用法の提供。【解決手段】変異型Ｆｃ断片は挿入を含み特定の配列の間にあり、又は特定の配列が欠失し、１０～２０アミノ酸の前記挿入は更に特定配列の間にあり、最初の４挿入されたアミノ酸の間と最後の４挿入されたアミノ酸の間に、其々少なくとも１つシステインを含み、前記変異型Ｆｃポリペプチドは挿入を含まないことを除いて、同じアミノ酸配列の対照Ｆｃポリペプチドよりも高い結合活性でヒト新生児Ｆｃ受容体（ｈＦｃＲｎ）に結合し、並びに前記変異型Ｆｃポリペプチドは、ｈＦｃＲｎ結合の結合活性がほとんど又は全く無く、検出される残留結合応答が前記対照Ｆｃポリペプチドを用いた検出よりも、０．１ナノメーター以下大きいヒトＩｇＧ１、ＩｇＧ２、ＩｇＧ３又はＩｇＧ４変異型Ｆｃ断片を含む前記変異型Ｆｃポリペプチド。【選択図】図１
