The present invention relates to a light-diffusing optical fiber probe which can safely and effectively treat a disease spread over a large area of tissue; and to a manufacturing method thereof. More specifically, a manufacturing method of a light-diffusing optical fiber probe uses a manufacturing apparatus of a light-diffusing optical fiber probe, which comprises an optical fiber holder, a processing control part, an optical fiber processing part, a side light sensor, a front light sensor, and a light energy supply part. The manufacturing method of the light-diffusing optical fiber probe comprises the following steps: a) mounting an optical fiber to be processed on the optical fiber holder; b) inputting a processing numerical value according to the range of light diffusion on the optical fiber to be processed, an energy distribution chart, the length of tissue to be treated, etc.; c) allowing the processing control part to output a processing control signal to control the optical fiber holder and the optical fiber processing part when the processing numerical value is inputted; d) allowing the optical fiber holder to actuate a rotating motor according to the processing control signal, thereby rotating the optical fiber mounted on the optical fiber holder; e) allowing the optical fiber processing part to be moved back and forth according to the processing control signal and process the side and front end of the optical fiber mounted on the optical fiber holder; f) transferring light energy to the optical fiber by the light energy supply part when the processing of the optical fiber is completed by the optical fiber processing part; g) measuring whether the light energy supplied from the light energy supply part is transferred to the side and front end of the optical fiber, via the side light sensor and the front light sensor; and h) comparing the measured intensity of light energy and the prestored energy distribution chart of the optical fiber to determine the supp
