The present application relates to arenaviruses that have a rearrangement of open reading frames (“ORFs”) in the genome. In particular, described herein are modified arenavirus genome segments that have been engineered to retain the viral ORF in a position that is distinct from the wild-type position of the ORF. Trisegmental arenavirus particles comprising one L segment and two S segments, or two L segments and one S segment are also described herein.
