摘要:
The invention relates to veterinary biotechnology, in particular, culturing animal cells and viruses, and can be used for optimising reproduction of viruses in a cell culture and for producing medicines for a specific immunoprotection against rabies. In order to improve the quality of the final product by increasing the yield of a viral antigen having an enhanced immunogenic activity, as well as to simplify the manufacturing technology of the rabies vaccine, in the method for producing rabies vaccine comprising cultivating BHK-21 transplantable cell culture, infecting the cells with rabies virus, replicating the virus, and inactivating the virus-containing suspension with ß-propiolactone followed by obtaining the final product, the cultivation of BHK-21 transplantable cell culture is performed for 48-72 hours to a concentration of (2.5-3.0)106 cells/ml followed by infecting the cells with rabies virus in a dose of 0.01-0.1 mMLD50/cell, maintaining at a temperature of 38.5-39.5°C for 60-90 minutes followed by diluting with a growth medium to a concentration of a cell sub-line of (0.5-0.6)106 cells/ml, and continuing the cultivation of the resulting suspension for 48-72 hours at a temperature of 37-38°C and a pH value of 7.1-7.3 subsequently, the virus-containing suspension is cooled to a temperature of 4-8°C at pH 7.8-8.0, and tris(hydroxymethyl)aminomethane and ethylenediaminotetraacetic acid disodium salt are added.
