PURPOSE: A method for enhancing long-term preservation efficiency of spermatogonial stem cells is provided to enhance the survival rate of the stem cells after thawing and to develop human infertility treatments. CONSTITUTION: A cryopreservation solution for spermatogonial stem cells contains 0.05-0.2M of trehalose. The solution is MEM-alpha containing 5-20% FBS and 5-20% DMSO. A method for cryopreservation of spermatogonial stem cells comprises: a step of treating the testis of mammals with enzyme to isolate single cell groups； a step of collecting spermatogonial stem cells from the single cell groups； a step of culturing the spermatogonial stem cells in vitro and collecting； and a step of performing cryopreservation of the solution.본 발명은 정원줄기세포의 장기간 보존 효율성을 증진시키는 방법에 관한 것이다. 보다 구체적으로 동결보존액에 트리할로오스 (trehalose)를 첨가한 동결보존법을 이용하여 정원줄기세포의 보존성을 증가시키는 방법을 제공하는 것이다.본 발명의 정원줄기세포 동결보존 기법을 따르면, 동결보존 및 해동 후 정원줄기세포의 생존율 증진을 확인할 수 있기 때문에 효율적인 동결보존을 통해 우수 종축의 형질 보존 연구 분야의 발전뿐 아니라 인간 불임치료와 같은 적극적인 활용법 개발에 획기적인 발전을 기대할 수 있다.
