[学术文献 ] Natural variation of an EF-hand Ca2+-binding-protein coding gene confers saline-alkaline tolerance in maize 进入全文

Sodium (Na+) toxicity is one of the major damages imposed on crops by saline-alkaline stress. Here we show that natural maize inbred lines display substantial variations in shoot Na+ contents and saline-alkaline (NaHCO3) tolerance, and reveal that ZmNSA1 (Na+ Content under Saline-Alkaline Condition) confers shoot Na+ variations under NaHCO3 condition by a genome-wide association study. Lacking of ZmNSA1 promotes shoot Na+ homeostasis by increasing root Na+ efflux. A naturally occurred 4-bp deletion decreases the translation efficiency of ZmNSA1 mRNA, thus promotes Na+ homeostasis. We further show that, under saline-alkaline condition, Ca2+ binds to the EF-hand domain of ZmNSA1 then triggers its degradation via 26S proteasome, which in turn increases the transcripts levels of PM-H+-ATPases (MHA2 and MHA4), and consequently enhances SOS1 Na+/H+ antiporter-mediated root Na+ efflux. Our studies reveal the mechanism of Ca2+-triggered saline-alkaline tolerance and provide an important gene target for breeding saline-alkaline tolerant maize varieties.

[学术文献 ] Calcium-activated 14-3-3 proteins as a molecular switch in salt stress tolerance 进入全文

Calcium is a universal secondary messenger that triggers many cellular responses. However, it is unclear how a calcium signal is coordinately decoded by different calcium sensors, which in turn regulate downstream targets to fulfill a specific physiological function. Here we show that SOS2-LIKE PROTEIN KINASE5 (PKS5) can negatively regulate the Salt-Overly-Sensitive signaling pathway in Arabidopsis. PKS5 can interact with and phosphorylate SOS2 at Ser294, promote the interaction between SOS2 and 14-3-3 proteins, and repress SOS2 activity. However, salt stress promotes an interaction between 14-3-3 proteins and PKS5, repressing its kinase activity and releasing inhibition of SOS2. We provide evidence that 14-3-3 proteins bind to Ca2+, and that Ca2+ modulates 14-3-3-dependent regulation of SOS2 and PKS5 kinase activity. Our results suggest that a salt-induced calcium signal is decoded by 14-3-3 and SOS3/SCaBP8 proteins, which selectively activate/inactivate the downstream protein kinases SOS2 and PKS5 to regulate Na+ homeostasis by coordinately mediating plasma membrane Na+/H+ antiporter and H+-ATPase activity.

[学术文献 ] Evaluation of maximum potential gene flow from herbicide resistant Brassica napus to its male sterile relatives under open and wind pollination conditions 进入全文

Pollen-mediated gene flow (PMGF) from genetically modified (GM) Brassica napus to its wild relatives by wind and insects is a major ecological concern in agricultural ecosystems. This study conducted is to estimate maximum potential gene flow and differentiate between wind- and bee-mediated gene flows from herbicide resistant (HR) B. napus to its closely-related male sterile (MS) relatives, B. napus, B. juncea and Raphanus sativus. Various markers, including pods formation in MS plants, herbicide resistance, and SSR markers, were used to identify the hybrids. Our results revealed the following: 1) maximum potential gene flow (a maximum % of the progeny of pollen recipient confirmed hybrid) to MS B. napus ranged from 32.48 to 0.30% and from 14.69 to 0.26% at 2–128?m from HR B. napus under open and wind pollination conditions, respectively, and to MS B. juncea ranged from 21.95 to 0.24% and from 6.16 to 0.16%, respectively; 2) estimates of honeybee-mediated gene flow decreased with increasing distance from HR B. napus and ranged from 17.78 to 0.03% at 2–128?m for MS B. napus and from 15.33 to 0.08% for MS B. juncea; 3) a small-scale donor plots would strongly favour insect over wind pollination; 4) no gene flow occurred from HR B. napus to MS R. sativus. Our approach and findings are helpful in understanding the relative contribution of wind and bees to gene flow and useful for estimating maximum potential gene flow and managing environmental risks associated with gene flow.

[学术文献 ] Dissolution and Regeneration in Films of Natural Luffa in 1-butyl-3-methylimidazolium Chloride 进入全文

Regenerated cellulose film was successfully prepared from natural luffa, a new cellulose raw material. A pretreatment of natural luffa was carried out by an alkali and hydrogen peroxide mixed solution. The dissolution process of the pretreated luffa in 1-butyl-3-methylimidazolium chloride ([BMIM]C1) was observed by polarized optical microscope. The structures and properties of the luffa films were characterized by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), thermogravimetric (TGA), and porosity measurements. The results showed that the luffa fibers were transformed to fibrils after the pretreatment. [BMIM]Cl was a good non-derivatizing solvent for the luffa cellulose. The solution conditions were 80°C and 10 h for a 15% solution. After being regenerated, as films, from the luffa/[BMIM]Cl solution, the crystalline structure of the luffa film was transformed completely from cellulose I to cellulose II. The film showed the strong characteristic functional groups of cellulose in FTIR results. The surface of the film was smooth with a compact structure. The porosity of the film was 66.2% and the average pore size was 17.8 nm. It was thermally stabile up to 280°C.

[学术文献 ] Esterification of Luffa with Acetic Anhydride under Microwave Radiation determined by FTIR 进入全文

Luffa, mainly made up of cellulose, hemicellulose, and lignin, is fruit of the sponge gourd. It has a special porous physical structure, which leads to a high adsorption property. In this study, luffa was pretreated in alkali/ethanol/water solution. XRD of luffa showed that after pretreatment, the cellulose structure in luffa changed from cellulose I to II, which implied that the crystal structure was destroyed to some extent and the accessibility to reaction was increased. The analysis by FT-IR indicated that the alkali treatment had little significant effect on luffa's chemical structure but can remove the lignin. The pretreated luffa was then esterified with acetic anhydride under microwave radiation, and the relative esterification degree was quantified by FT-IR using an improved internal standard method. The results showed that microwave radiation could rapidly increase the esterification degree in the reaction process. The esterification degree of luffa esterified 20 min under microwave radiation was nearly 25 times that esterified 90 min under oil heat. The esterification degree initially increased quickly with radiation time and trended to become constant after 15 min radiation.

SUAEDA SALSA L. is a typical euhalophyte and is widely distributed throughout the world. Suaeda plants are important halophyte resources, and the physiological and biochemical characteristics of their various organsand their response to salt stress have been intensively studied. Leaf succulence, intracellular ion localization, increased osmotic regulation and enhanced antioxidant capacities are important responses for Suaeda plants to adapt to salt stress. Among these responses, scavenging of reactive oxygen species (ROS) is an important mechanism for plants to withstand oxidative stress and improve salt tolerance. The generation and scavenging pathways of ROS, as well as the expression of scavenging enzymes change under salt stress. This article reviews the antioxidant system constitute of S. salsa, and the mechanisms by which S. salsaantioxidant capacity is improved for salt tolerance. In addition, the differences between types of antioxidant mechanisms in S. salsaare reviewed, thereby revealing the adaptation mechanisms of Suaeda to different habitats. The review provides important clues for the comprehensive understanding of the salt tolerance mechanisms of halophytes.
