1429096 Antibiotic 60-6 SHIONOGI & CO Ltd 24 April 1974 [25 April 1973] 17941/74 Heading C2A An antibiotic, designated 60-6, which antibiotic is a colourless amphoteric powder which shows no definite M.Pt. but progressively decomposes over 140?? C.; which contains C, H, N and O in the following proportions by wt.: which has an optical rotation of [α]23??0 D + 19??8 ?? 3??5?? (C=0??172% in dimethylformamide); which has a M.Wt. of 1600 and a neutralization equivalent of 1517; which forms aspartic acid, threonine, serine, valine, alloisoleucine, tryptophan,γ-hydroxylysine, ammonia and a variety of fatty acids of 12-16 carbon atoms upon acid hydrolysis; which has ultraviolet absorption bands (in methanol) at 275, 282 and 290??5 m??; which has a solubility as defined; which exhibits positive ninhydrin and Dragendorff colour reactions; which exhibits behaviour on t.l.c. as specified; and which has an infra-red absorption spectrum as shown in the accompanying drawing. The antibiotic may be prepared by cultivating a suitable strain of Bacillus cereus in an aqueous nutrient medium under aerobic conditions. A suitable strain is Bacillus cereus No. 60-6, ATCC No. 21929. Preferred cultivation conditions are a temperature of 20-37?? C., submerged aerobic conditions, a nutrient medium pH of 7-8, and the optional presence of a buffering agent and an anti-foaming agent. The cultivation proceeds preferably for 20-100 hours, after which the antibiotic may be isolated by filtering the broth and extracting both cells and filtrate with a suitable solvent. There may be prepared, from the antibiotic, salts such as an acid addition salt, an ammonium salt or a pharmaceutically acceptable salt. The antibiotic may be formed into a pharmaceutical or disinfectant composition by admixture with a suitable diluent, carrier or excipient.
