The present invention relates to a method for preparing a coffee extract by bioconversion which prepares a coffee extract by the following steps: grinding raw coffee beans based on the digestive system and digestive enzymes of a civet cat； preparing a mixture by adding malt, papain, and a castor bean extract to the ground raw coffee beans and making the same react with each other； inoculating the mixture with microorganisms； and cultivating the resultant product. The coffee extract, prepared by the preparation method of the present invention: is extracted by a simple extraction method； is prepared by mixing the mixture including raw coffee beans and the like with the inoculated microorganisms to be newly bioconverted, and thus has high contents of various components, such as polyphenol and flavonoid, while the conventional luwak coffee obtained by the civet cat has low contents of components since the various components are absorbed or consumed in the intestine of the cat； has higher amino acid content, which determines the characteristics of the luwak coffee, than the conventional luwak coffee obtained by digestion products of the civet cat； and causes the unique taste of coffee.본 발명은 사양 고양이의 소화기관과 소화 효소의 기능을 바탕으로 하여 분쇄한 생두에 엿기름, 파파인, 아주까리 추출물을 첨가하여 반응시킨 혼합물에 미생물을 접종하고 배양하여 생물전환 커피 추출물을 제조함으로써, 커피의 추출방법이 매우 간단할 뿐만 아니라 종래 사양 고양이에 의해 얻어지는 루왁 커피의 경우에는 고양이의 장내에서 각종의 성분들이 흡수 또는 소모되어 성분들의 함유량이 매우 낮은데 비해 본 발명의 경우에는 생두 등의 혼합물과 접종된 미생물이 어우러져 새롭게 생물 전환되어 생성됨으로 인해 폴리페놀, 플라보노이드와 같은 각종 성분들의 함유량이 매우 높고 그리고 루왁 커피의 특성을 확정 짖는 아미노산 함량이 종래의 사양 고양이의 소화 산물에 의해 얻은 루왁 커피에 비해 매우 높으면서 커피의 특이한 취향을 유발할 수 있는 것에 장점이 있다.
