PURPOSE: A manufacturing method of a bilayer scaffold for artificial skin tissues is provided to manufacture the bilayer scaffold by fusing a porous polymer hydrogel which is prepared by a casting process of a synthetic polymer and a freeze-drying process. CONSTITUTION: A manufacturing method of a bilayer scaffold for artificial skin tissues comprises the following steps. An upper layer is manufactured by means of an electrospinning process. A lower layer is manufactured by means of casting, pre-freezing, and free-drying processes. An upper membrane layer is first processed by the electrospinning process. A lower membrane is formed by casting chitosan/gelatin solutions in a polystyrene mold. After casting the chitosan/gelatin solutions, a pre-freezing process to facilitate the formation of ice crystals and a free-drying process to form a porous hydrogel network are performed.인공피부조직용 이중층 지지체의 제조방법이 개시되어 있다. 본 발명은, 폴리카프로락톤(polycaprolactone : PCL)과 폴리락토코글리코라이드(poly(lacto-co-glycolide) : PLGA)를 용매(tetrahydrofuran, dimethylformamide, methylene chloride)로 용해시켜 혼합한 후, 혼합된 폴리머를 전기방사시켜 섬유매트를 제조하는 단계； 키토산과 젤라틴을 각각 1%(v/v) 아세트산과 탈이온화 된 물로 용해시켜 혼합시킨 후 글루타르알데히드로 가교시켜서, 키토산과 젤라틴 하이드로겔 복합체를 제조하는 단계； 조직배양판 내에 상기 섬유매트를 배치시키고, 그 위에 상기 키토산과 젤라틴 하이드로겔 복합체 용액을 투입한 후에, 실온으로 유지시켜서 상기 복합체 용액을 고화시키는 단계； 빙정을 형성하기 위하여 상기 섬유매트와 고화된 복합체 용액을 포함하는 조직배양판을 동결시키는 단계； 및 상기 동결이 완료된 조직배양판을 동결 건조시키는 단계；를 포함하는 것을 특징으로 한다.
