A complex containing a non-enzymatically self-cleaving fragment covalently linked to a drug, said self-cleaving fragment containing the general structure A-B-, where A is an amino acid or hydroxy acid； B is an N-alkylated amino acid； wherein said self-cleaving fragment is bound to said drug by forming an amide bond between B and the amine in the composition of said drug, wherein the half-life of chemical cleavage of (t) AB from said drug is at least about 1 hour to about 1 week in standard PBS solution under standard physiological conditions. 2.
