The invention relates to a process of preliminary selection (screening) for the resistance of maize lines as regards the contamination with aflatoxins during vegetation, intended to be used in the improvement of maize crops. According to the invention, the process includes the following stages: finely grinding the grains to be analyzed, total extraction of proteins in citrate buffer at a pH = 5.5, for 1 h, in a maize grains : buffer ratio of 1 : 5, separating the vegetable material extract by centrifugation at 6000 x g, for 20 min, sterilizing the grain extract by ultrafiltration, aseptically depositing 0.02 ml of filtration-sterilized extract at the surface of a 1 ml well in a plate with 24 wells, containing 0.5 ml of a gelled culture medium, axenic inoculation of the gelled medium containing the protein extract with 0.01 ml of aflatoxigenic fungi spore suspension, incubation for 3 days, at 25°C, followed by examination of the intensity of fluorescence formed at an excitation wavelength of 350...360 nm and a reading wavelength of 450...460 nm.Prezenta invenţie se referă la un procedeu de selecţie preliminară (screening) a rezistenţei liniilor de porumb din punctul de vedere al rezistenţei la contaminarea cu aflatoxine în timpul vegetaţiei, destinat utilizării în activitatea de ameliorare a culturilor de porumb. Procedeul conform invenţiei include etapele de măcinare fină a boabelor analizate, extracţia totală a proteinelor în tampon citratH = 5,5, în raport de 1 parte boabe de porumb la 5 părţi tampon, timp de 1 h, separarea extractului de material vegetal prin centrifugare timp de 20 min, la 6000 x g, sterilizarea extractului de boabe prin ultrafiltrare, depunerea aseptică a 0,02 ml extract sterilizat prin filtrare la suprafaţa unui godeu de 1 ml dintr-o placă de 24 godeuri, în care se află 0,5 ml dintr-un mediu de cultură gelificat, inocularea axenică a mediului gelificat ce conţine extractul proteic cu 0,01 ml suspensie de spori de ciuperci aflatoxigene, in
