Disclosed herein are a modified lysosomal protein, methods for preparing a modified lysosomal protein and therapeutic use of such a modified protein. Further disclosed herein is a method of treating a mammal afflicted with a lysosomal storage disease.In particular, the present disclosure relates to a method of preparing a modified lysosomal protein, said method comprising reacting a glycosylated lysosomal protein with an alkali metal periodate and reacting said lysosomal protein with an alkali metal borohydride for a time period of no more than 2 h, thereby modifying glycan moieties of the lysosomal protein and reducing the activity of the lysosomal protein with respect to glycan recognition receptors.L'invention concerne une protéine lysosomale modifiée, des procédés de préparation d'une protéine lysosomale modifiée et l'utilisation thérapeutique d'une telle protéine modifiée. L'invention concerne également une méthode de traitement d'un mammifère atteint d'une maladie lysosomale. En particulier, la présente invention concerne un procédé de préparation d'une protéine lysosomale modifiée, ledit procédé consistant à faire réagir une protéine lysosomale glycosylée avec un periodate de métal alcalin et à faire réagir ladite protéine lysosomale avec un borohydrure de métal alcalin pendant une durée n'excédant pas 2 h, ce qui permet de modifier des fragments de glycane de la protéine lysosomale et de réduire l'activité de la protéine lysosomale par rapport aux récepteurs de reconnaissance du glycane.
