PROBLEM TO BE SOLVED: To provide a method for preparing a monolayer liposome composition of a polyene antibiotic.SOLUTION: The present invention provides a method that comprises the steps of: forming a soluble complex of a polyene antibiotic and phospholipid in an acidified organic solvent and maintaining the soluble complex at a pH of 4.5 or less； removing the organic solvent to form a dried complex and hydrating the dried complex with an aqueous buffer solution to form a dispersion of monolayer liposomes containing the polyol antibiotic from a mixture resulted from the hydration； and subjecting the dispersion to filter sterilization.
