Provided are pharmaceutical compositions and methods for immunotherapy. The pharmaceutical composition comprises a genetically engineered bacterium which expresses an antigen associated with human disease, preferably a tumor antigen specific for more than one patient as a polytope. This bacterium has lipopolysaccharides engineered to express endotoxin at low levels that are insufficient to induce CD14-mediated sepsis or the patient's own endosymbiont. The genetically engineered bacteria can be administered to a patient systemically or locally to induce a tumor-specific immune response.
