The present invention relates to a stem cell and, more specifically, a composition for inhibiting differentiation or aging of mesenchymal stem cells, and a screening method thereof where IL-6 which is a causing factor to make stem cells aging and lose pluripotency and Runx2/Dlx5 induced by the IL-6 are newly discovered, and the inhibition of expressing the IL-6 and Runx2/Dlx5 prevents aging of stem cell, while stem cells are long cultured, and the stem cell maintains its pluripotency. In the present invention, problems that stem cells are aging and are intensively turned into bone differentiation are found in a long term culture of stem cells, which passed many passages in vitro to obtain treating effective amount of stem cells, and are fundamentally resolved, thereby efficiently using stem cells as treatment resources whose scarcity value is high.COPYRIGHT KIPO 2015본 발명은 줄기세포, 보다 구체적으로는 중간엽 줄기세포의 분화 또는 노화 억제용 조성물 및 이의 스크리닝 방법에 관한 것이다.본 발명은 줄기세포의 노화 및 다분화능 상실의 원인 인자로서 IL-6 및이에 의해 발현이 유도되는 Runx2/Dlx5를 새롭게 발굴하였으며, 이들의 발현 억제를 통해 장기간의 배양기간에도 줄기세포의 노화가 억제되고 다분화능이 유지될 수 있음을 제안한다. 본 발명은 줄기세포의 치료적 유효량을 수득하기 위해 인 비트로에서 여러 계대를 거치면서 장기 배양을 할 때 나타나는 노화 및 골 계열로의 분화 치중의 문제를 근본적으로 해결함으로써 희소가치가 높은 치료자원으로서의 줄기세포를 보다 효율적으로 이용할 수 있도록 한다.
