This invention provides flavivirus having a viral genome modified by the introduction of one or more mutations. The flavivirus may be attenuated and may have a phenotype that is temperature sensitivity, host-cell restriction or host-cell adaptation for improved replication. A menu of mutations was developed that is useful in fine- tuning attenuation and growth characteristics of dengue virus vaccines.
