The present invention is directed to nanostructured (nanoparticulated) Sildenafil base, its pharmaceutically acceptable salts and co-crystals, compositions containing them, process for the preparation thereof and pharmaceutical compositions containing them. The nanoparticles of Sildenafil base, its pharmaceutically acceptable salts and co-crystals, compositions containing them according to the invention have an average particle size of less than about 500 nm. Sildenafil citrate is inhibiting cGMP specific phosphodiesterase type 5 (PDEV), an enzyme that regulates blood flow in the penis. The compositions of the invention are useful in the treatment of male or female sexual dysfunction and pulmonary arterial hypertension (PAH).L'invention concerne du Sildénafil base nanostructuré (nanoparticulaire), des sels et des cocristaux pharmaceutiquement acceptables de ce composé, des compositions contenant celui-ci, un procédé de préparation de ce composé et des compositions pharmaceutiques contenant ceux-ci. Les nanoparticules de Sildénafil base, les sels et cocristaux pharmaceutiquement acceptables de ce composé, les compositions contenant ceux-ci, selon l'invention, présentent une dimension moyenne des particules inférieure à environ 500 nm. Le Sildénafil citrate inhibe la phosphodiestérase de type 5 (PDEV) spécifique de cGMP, une enzyme qui régule le flux sanguin dans le pénis. Les compositions de l'invention sont utiles pour traiter un dysfonctionnement sexuel chez l'homme ou la femme ainsi que l'hypertension artérielle pulmonaire (HTAP).
