An anesthetic composition is provided for topical administration of an amide-type anesthetic to a subject in need thereof. The anesthetic composition has a multi-layered vesicle with a captured amide-type anesthetic prepared by hydrating a highly captured lipid construct comprising an amide-type anesthetic and lipid mixture with a buffered aqueous solution having a pH greater than 5.5. Also provided is a method of making an anesthetic composition that uses a simpler and more feasible process for large scale manufacturing and provides a high molar ratio of amide-type anesthetic to phospholipid content compared to the prior art.
