PROBLEM TO BE SOLVED: To provide a mono-pegylated (pegylated PEG), and di-pegylated interleukin 10 (IL10) composition, and to provide a method for using the same.SOLUTION: The present invention provides a method of producing a mixture of mono- and di-pegylated IL-10, wherein at least one PEG molecule is covalently attached to at least one amino acid residue of at least one subunit of IL-10, comprising: a) reacting 1 mg/ml to 12 mg/ml of IL-10 protein with an activated PEG-linker such that the IL-10 to PEG-linker ratio is 1:1 to 1:7.7, in the presence of 0.75 mM to 35 mM of a reducing agent, at a pH of about 5.0 to 7.4 and a temperature of 5°C to 30°C, for 12-15 hours； and b) purifying the mixture of mono- and di-pegylated IL-10.【課題】モノＰＥＧ化（ｐｅｇｙｌａｔｅｄ ＰＥＧ）およびジＰＥＧ化インターロイキン１０（ＩＬ１０）組成物及びその使用方法の提供。【解決手段】ａ）０．７５ｍＭ～３５ｍＭの還元剤の存在下、約５．０～７．４のｐＨ及び５℃～３０℃の温度で１２～１５時間にわたり、１ｍｇ／ｍｌ～１２ｍｇ／ｍｌのＩＬ１０タンパク質を活性化ＰＥＧ－リンカーと反応させ、この場合、ＩＬ１０対ＰＥＧ－リンカーの比が１：１～１：７．７となるようにし、ｂ）モノ－及びジ－ＰＥＧ化ＩＬ１０の混合物を精製することを含む、少なくとも１つのＰＥＧ分子がＩＬ１０の少なくとも１つのサブユニットの少なくとも１つのアミノ酸残基に共有結合しているモノ－及びジ－ＰＥＧ化ＩＬ１０の混合物の製造方法。【選択図】図１
