The present invention relates to the simple and cost effective process for preparation of pharmacopoeial grade Insulin Glargine from a bacterial fermentation process followed by isolation of IBs, refolding, enzymatic conversion and chromatographic purification. Pure Insulin Glargine is lyophilized or crystallized to obtain pharmacopoeial grade Insulin Glargine. The invention also relates to the process of enzymatic conversion of purified Proinsulin Glargine using trypsin in the chromatographic column elute, wherein the ratio of trypsin to purified Proinsulin Glargine is 1:500 to 1:2000.
