The present invention provides compositions and methods for down-regulating major histocompatibility class I molecule cell surface expression, as well as isolated T cells (eg, genetically modified antigen-specific such as chimeric antigen receptor T (CAR-T) cells. Use of such compositions and methods for improving the functional activity of (T cells) is provided. In particular, the invention provides methods and compositions for supporting the therapeutic efficacy of CAR-T cells.
