A trifunctional molecule is provided that includes a target-specific ligand, a ligand that binds to a protein that associates with a TCR complex, and a T cell receptor signaling domain polypeptide. Manipulating T cells with this novel receptor results in antigen-specific activation of numerous T cell functions including cytokine production, degranulation and cytolysis.
