The present disclosure involves compositions and methods for treating brain cancers having mutations in the retinoblastoma (Rb) pathway using an oncolytic adenovirus comprising an alteration in the Rb binding site of E1A, and a targeting motif inserted in the Ad fiber protein. The adenovirus is able to kill the tumor cells without harming cells with a wild-type retinoblastoma pathway.La présente invention concerne des compositions et des méthodes pour le traitement de cancers du cerveau présentant des mutations au niveau de la voie du rétinoblastome (Rb) à l'aide d'un adénovirus oncolytique comprenant une modification du site de liaison à Rb d'El A, et un motif de ciblage inséré dans la protéine fibreuse Ad. L'adénovirus peut tuer les cellules cancéreuses sans endommager les cellules présentant une voie du rétinoblastome de type sauvage.
