Allylomycin analogs that can have broad spectrum biological activity are provided. Resistance to the antibacterial biological activity of the natural product allylomycin in a range of pathogenic bacterial species was found to depend on a single amino acid mutation at a defined position of the bacterial signal peptidase (SPase), where the presence of a proline residue Confers allylomycin resistance. Provided herein are allylomycin analogs that can overcome that resistance and provide a broader range of antimicrobial bioactivity than can be provided by the natural product allylomycin, such as allylomycin A2. Methods are provided for determining whether a bacterial strain is sensitive to narrow-area allylomycin antibiotics or whether a broad analog is required for treatment.
