The present invention relates to methods for treating mitochondrial genetic disorders. We studied with primary fibroblasts from patient and control individuals and collected protein lysates for Western blots. Importantly, we observed that mitochondrial gene abnormalities showed a significant increase in phosphorylation of ribosomal protein S6 (pS6) compared to control fibroblasts, which was excessive. Shown is activated mTOR signaling. Patients and control cells with mitochondrial abnormalities were treated with DMSO or BYL719 for 48 hours. All strains of patients with mitochondrial disease had reduced membrane potential as determined by tetramethylrhodamine ethyl ester (TMRE) staining intensity, as well as abnormal morphology, fragmentation, and depolarization of mitochondria ( Low) indicating the presence of mitochondria.
