A non-naturally occurring polypeptide or isolated polypeptide having a ratio of at least 1.3 of an activity in an assay at a first pH in the presence of at least one species having a molecular weight of less than 900 a.m.u. and a pKa up to 4 pH units away from said first pH, to an activity in an assay at a second pH in the presence of the same at least one species. The species has a pKa beween said first pH and said second pH and can be a small molecule. Also disclosed are pharmaceutical formulations including the polypeptide and uses thereof. Methods of producing conditionally active polypeptides are also disclosed.La présente invention concerne un polypeptide non d'origine naturelle ou un polypeptide isolé présentant un rapport d'au moins 1,3 d'une activité dans un essai sous un premier pH en présence d'au moins une espèce présentant un poids moléculaire inférieur à 900 a.m.u. et un pKa allant jusqu'à 4 unités de pH éloigné dudit premier pH, jusqu'à une activité dans un essai sous un second pH en présence de la même espèce. L'espèce présente un pKa compris entre ledit premier pH et ledit second pH et peut être une petite molécule. Sont également décrites des formulations pharmaceutiques comprenant le polypeptide et leurs utilisations.
