The present invention discloses latent thioester functionalities attached to the C-terminus of a first polypeptide, or a first fragment thereof having a Cys residue at its N-terminus, and a process using this functionality for the preparation of polypeptide thioesters, in particular of ubiquitin thioesters, this process comprising preparing a polypeptide or a fragment thereof, being attached to a latent thioester functionality, which can then be ligated with a second polypeptide fragment, followed by selective activation of the latent thioester functionality group, to provide the requested polypeptide thioester. There are also provided the polypeptides obtained by this method, specific unnatural amino acids useful to be incorporated within the polypeptide thioesters, and kits for preparing them.La présente invention concerne des fonctionnalités thioester latentes attachées à la terminaison C dun premier polypeptide, ou à un premier fragment de ce dernier ayant un reste Cys au niveau de sa terminaison N, ainsi quun procédé dans lequel on utilise cette fonctionnalité pour préparer des thioesters polypeptidiques, notamment des thioesters dubiquitine. Le procédé selon linvention consiste à préparer un polypeptide ou un fragment de polypeptide, attaché à une fonctionnalité thioester latente, qui peut ensuite être lié à un second fragment polypeptidique, puis à activer sélectivement le groupe à fonctionnalité thioester latente, pour produire le thioester polypeptidique demandé. Linvention porte également sur les polypeptides obtenus au moyen dudit procédé, des acides aminés spécifiques utiles à incorporer dans les thioesters polypeptidiques et des trousses pour les préparer.
