The present invention relates to a pharmaceutical composition for preventing or treating bone metabolic diseases comprising a lysyl-tRNA synthetase (KRS) inhibitor as an effective component. Specifically, a material for inhibiting KRS of the present invention inhibits the formation of osteoclasts, inhibits the expression of transcription factors expressed by the RANK signaling pathway in osteoclasts, and inhibits the activation of the MAPK signaling pathway, and thus can be usefully used for the treatment of bone metabolic diseases caused by the formation of osteoclasts.본 발명은 라이실-tRNA 합성효소(lysyl-tRNA synthetase, KRS) 억제제를 유효성분으로 함유하는 골대사성 질환 예방 또는 치료용 약학적 조성물에 관한 것이다. 구체적으로, 본 발명의 KRS를 억제하는 물질은 파골세포의 형성을 억제하고, 파골세포에서 RANK의 신호전달기전에 의해 발현되는 전사인자의 발현을 억제시킬뿐만 아니라, MAPK 신호전달 기전의 활성화를 억제함으로써, 상기 KRS를 억제하는 물질은 파골세포의 형성에 의해 발생하는 골대사성 질환의 치료에 유용하게 사용될 수 있다.
