Described herein are methods of identifying immunologically protective neo-epitopes from the cancer tissue DNA of cancer patients using biophysical principles as well as bioinformatics techniques. The identification of immunologically protective neo-epitopes provides pharmaceutical compositions with a limited number of tumor-specific peptides suitable for personalized genomics-driven immunotherapy of human cancer. Specifically disclosed herein is a method of using the conformational stability of an epitope in an MHC protein -binding groove to predict immunogenicity of peptides in a putative neo-peptide set from a tumor from a cancer patient. Pharmaceutical compositions and methods of administration are also included.L'invention concerne des méthodes d'identification de néo-épitopes immunologiquement protecteurs présents dans l'ADN du tissu tumoral de patients cancéreux faisant appel à des principes biophysiques, ainsi qu'à des techniques bio-informatiques. L'identification de néo-épitopes immunologiquement protecteurs permet d'obtenir des compositions pharmaceutiques comportant un nombre limité de peptides spécifiques d'une tumeur appropriés à l'immunothérapie anticancéreuse personnalisée fondée sur la génomique chez l'être humain. L''invention concerne, plus précisément, un procédé d'utilisation de la stabilité de conformation d'un épitope dans un sillon de liaison aux protéines du CMH pour prédire l'immunogénicité de peptides au sein d'un ensemble de néo-peptides putatifs provenant d'une tumeur d'un patient cancéreux L'invention concerne également des compositions pharmaceutiques et leurs procédés d'administration.
