Disclosed is a humanised antibody or fragment thereof capable of binding to GD2, wherein the antibody or fragment thereof comprises the amino acid sequences of CDR1, CDR2 and CDR3 of the murine antibody m3F8 variable light chain and the amino acid sequences of CDR1, CDR2 and CDR3 of the murine antibody m3F8 variable heavy chain, and wherein: a) the humanised antibody or fragment thereof confers on an IgG1 comprising the antibody or fragment thereof a capability of mediating ADCC activity against neuroblastoma LAN-1-PBMC at least about 10 fold or greater than that of m3F8 or b) the humanised antibody or fragment thereof confers on an IgG4 comprising the antibody or fragment thereof a capability of blocking pain in a rat allodynia model.
