Anti-VISTA monoclonal antibodies and related compositions are provided, which may be used in any of a variety of therapeutic methods for the treatment of various inflammatory, neoplastic, and infectious diseases. A heavy chain variable region that binds to VISTA and comprises (i) VHCDR1, VHCDR2, and VHCDR3 of SEQ ID NOs: 3-5, 11-13, 19-21, 2-29, 35-37, or 43-45. (Ii) Containing light chain variable regions comprising VLCDR1, VLCDR2, and VLCDR3 of SEQ ID NOs: 6-8, 14-16, 22-24, 30-32, 38-40, or 46-48, respectively. Heavy and light chains that are identical to the heavy and light chain variable regions of (i) and (ii), except for isolated antibodies or antigen-binding fragments thereof, or up to eight amino acid substitutions in the CDR regions.
