PROBLEM TO BE SOLVED: To efficiently and surely grow and culture Volvariella speciosa sing. var. volvacea and its spawn. SOLUTION: This method for culturing a spawn of Volvariella speciosa sing. var. volvacea comprises a first step for supplying sufficient water to straws 8 in a state of the original shape and removing lye, a second step for cutting the straws 8 into chips of a prescribed size, formulating and mixing a rice bran, cottonseeds, cattle feces and slaked lime therewith and producing a culture medium material 9, a third step for pressing and stuffing the culture medium material 9 into a bottle, closing an inlet of the bottle with a cotton plug, covering the top of the cotton plug with a film, sealing the film with a rubber band and forming a culture medium, a fourth step for housing the bottle in a sterilizing container (a sterilizing chamber), sealing steam in the interior of the sterilizing container, pressurizing the sterilizing container and keeping the interior of the sterilizing container at about 120 deg.C, a fifth step for holding the bottle in the sterilizing container for a prescribed time, a sixth step for stopping the pressurization and steam sterilization and allowing the bottle to stand for a prescribed time, a seventh step for repressurizing the interior of the sterilizing container, sealing steam therein and keeping the interior of the sterilizing container at about 120 deg.C, an eighth step for holding the bottle in the sterilizing container for 2-3 h, a ninth step for forming a hole in the center of the culture medium material 9 stuffed in the bottle and inoculating a spawn of the Volvariella speciosa sing. var. volvacea after reducing the temperature of the culture medium material 9 to a prescribed temperature and a tenth step for keeping the temperature of the culture medium material 9 at a prescribed temperature in a dark room.
