Disclosed is an amanitin derivative that is conjugated to a targeted antibody to form an ADC (antibody drug conjugate). The present disclosure provides improved amatoxin derivatives for use in ADC (antibody drug conjugate) structures. More specifically, the present disclosure provides an antibody drug conjugate (ADC) having the structure of Formula I or a pharmaceutically acceptable salt thereof.
