Disclosed is a method for production of recombinant human alpha-galactosidase A (rh alpha-Gal A) in a large scale, with a high purity. The method comprises the steps of (a) culturing rh alpha-Gal A-producing mammalian cells in a serum-free medium, (b) collecting culture supernatant, (c) subjecting the culture supernatant to anion-exchange column chromatography, (d) to hydrophobic column chromatography, (e) to a column chromatography employing as solid phase a material having affinity for phosphate group, (f) to cation-exchange column chromatography, (g) to dye-affinity column chromatography, and (h) to gel filtration column chromatography, in the order.Linvention concerne un procédé de production à grande échelle, avec une haute pureté, dalpha-galactosidase A humaine de recombinaison (rh alpha-Gal A). Le procédé comprend les étapes consistant, dans lordre, (a) à cultiver des cellules mammifères produisant la rh alpha-Gal A dans un milieu exempt de sérum, (b) à recueillir le surnageant de la culture, (c) à soumettre le surnageant de la culture à une chromatographie sur colonne à échange de cations, (d) à une chromatographie sur colonne hydrophobe, (e) à une chromatographie sur colonne utilisant en tant que phase solide, un matériau ayant une affinité pour le groupe phosphate, (f) à une chromatographie sur colonne à échange de cations, (g) à une chromatographie sur colonne à affinité de colorant et (h) à une chromatographie sur colonne de filtration sur gel.
